Understanding stem cell states: naïve to primed pluripotency in rodents and humans

چکیده
Preface The molecular mechanisms and signalling pathways that regulate the in vitro preservation of distinct pluripotent stem cell configurations, and their induction in somatic cells via direct reprogramming approaches, continue to constitute a highly exciting area of research. In this Review, we provide an integrative synthesis on recent discoveries related to isolating unique naïve and primed pluripotent stem cell states with altered functional and molecular characteristics, and from different species. We overview pathways underlying pluripotent state transitions and interconversion in vitro and in vivo. We conclude by highlighting unresolved key questions, future directions and potential novel applications of such pluripotent cell states.

Spermatogonia stem cells: A new pluripotent source for repairment in regenerative medicine
Recently new reports have proved the pluripotency of spermatogonial stem cells (SSCs) derived from male gonad. This pluripotent stem cells resembled Embryonic stem cells recognized as Embryonic Stem like cells (ES like cells). ES like cells forms sharp edge colonies that are immunopositive to pluripotency markers and have differentiation capacity to Ectodermal, Mesodermal and Endodermal layers....
متن کاملWnt/β-catenin signaling promotes self-renewal and inhibits the primed state transition in naïve human embryonic stem cells.
In both mice and humans, pluripotent stem cells (PSCs) exist in at least two distinct states of pluripotency, known as the naïve and primed states. Our understanding of the intrinsic and extrinsic factors that enable PSCs to self-renew and to transition between different pluripotent states is important for understanding early development. In mouse embryonic stem cells (mESCs), Wnt proteins stim...
متن کاملNuclear Architecture and Epigenetics of Lineage Choice
Differentiation is an epigenetic process which is installed by changes of transcriptional programs over successive cellular divisions. A number of studies have reported the effects of biochemical modifications of chromatin (DNA and chromatin proteins) on the regulation of transcription. Although, these studies are able to explain how transcription of a given gene is regulated (toward activation...
متن کاملComparative computational analysis of pluripotency in human and mouse stem cells
Pluripotent cells can be subdivided into two distinct states, the naïve and the primed state, the latter being further advanced on the path of differentiation. There are substantial differences in the regulation of pluripotency between human and mouse, and in humans only stem cells that resemble the primed state in mouse are readily available. Reprogramming of human stem cells into a more naïve...
متن کاملA Simple Method to Identify Kinases That Regulate Embryonic Stem Cell Pluripotency by High-throughput Inhibitor Screening
Embryonic stem cells (ESCs) can self-renew or differentiate into all cell types, a phenomenon known as pluripotency. Distinct pluripotent states have been described, termed "naïve" and "primed" pluripotency. The mechanisms that control naïve-primed transition are poorly understood. In particular, we remain poorly informed about protein kinases that specify naïve and primed pluripotent states, d...
